چکیده
Abstract BACKGROUND: In vitroexperiments in which HIV-infected CD4 +T cells are co-cultured with natural killer (NK) yield incomplete elimination of the infected cells. We therefore hypothesize that cell-intrinsic pathways differentially regulated to mediate resistance NK cell-mediated killing. METHODS: Mock-infected and HIV-89.6-infected from 6 HIV −donors were overnight +/− autologous Following co-culture, stained for surface exposed envelope protein using fluorescently conjugated antibodies. Fluorescence activated cell sorting was then used isolate uninfected cells, followed by bulk RNA-sequencing. Transcripts versus without compared. RESULTS: RNA-seq analysis survived co-culture revealed dozens expressed genes infected, but not This included upregulation interferon-stimulated genes, including PD-L1 (p adj=3×10 −6), NF-kB-related genes. Additional upregulated surviving c-Fos adj=1.9×10 −5), c-Jun adj=4.9×10 −8), cathepsin L adj=5×10 −3). CONCLUSIONS: NF-kB downregulated potentially through Vpu activity, normalized may be due targeting Vpu-mediated HLA-C downregulation. Deconvolution relative contributions specific genes/proteins target will reveal novel targets development therapeutics eliminate reservoir.

Small CD4 Mimetics Prevent HIV-1 Uninfected Bystander CD4 + T Cell Killing Mediated by Antibody-dependent Cell-mediated Cytotoxicity
Human immunodeficiency virus type 1 (HIV-1) infection causes a progressive depletion of CD4 + T cells. Despite its importance for HIV-1 pathogenesis, the precise mechanisms underlying CD4 + T-cell depletion remain incompletely understood. Here we make the surprising observation that antibody-dependent cell-mediated cytotoxicity (ADCC) mediates the death of uninfected bystander CD4 + T cells in ...
متن کاملHistone Deacetylase Inhibitors Enhance CD4 T Cell Susceptibility to NK Cell Killing but Reduce NK Cell Function
In the search for a cure for HIV-1 infection, histone deacetylase inhibitors (HDACi) are being investigated as activators of latently infected CD4 T cells to promote their targeting by cytotoxic T-lymphocytes (CTL). However, HDACi may also inhibit CTL function, suggesting different immunotherapy approaches may need to be explored. Here, we study the impact of different HDACi on both Natural Kil...
متن کاملComparative Efficiency of HIV-1-Infected T Cell Killing by NK Cells, Monocytes and Neutrophils
HIV-1 infected cells are eliminated in infected individuals by a variety of cellular mechanisms, the best characterized of which are cytotoxic T cell and NK cell-mediated killing. An additional antiviral mechanism is antibody-dependent cellular cytotoxicity. Here we use primary CD4(+) T cells infected with the BaL clone of HIV-1 as target cells and autologous NK cells, monocytes, and neutrophil...
متن کاملTryptophan Biosynthesis Protects Mycobacteria from CD4 T-Cell-Mediated Killing
Bacteria that cause disease rely on their ability to counteract and overcome host defenses. Here, we present a genome-scale study of Mycobacterium tuberculosis (Mtb) that uncovers the bacterial determinants of surviving host immunity, sets of genes we term "counteractomes." Through this analysis, we found that CD4 T cells attempt to contain Mtb growth by starving it of tryptophan--a mechanism t...
متن کاملImpaired CD4 T Cell Memory Response to Streptococcus pneumoniae Precedes CD4 T Cell Depletion in HIV-Infected Malawian Adults
OBJECTIVE Invasive pneumococcal disease (IPD) is a leading cause of morbidity and mortality in HIV-infected African adults. CD4 T cell depletion may partially explain this high disease burden but those with relatively preserved T cell numbers are still at increased risk of IPD. This study evaluated the extent of pneumococcal-specific T cell memory dysfunction in asymptomatic HIV infection early...
